Avana is a prescription medication containing the active ingredient avanafil, a potent and selective phosphodiesterase type 5 (PDE5) inhibitor specifically formulated for the treatment of erectile dysfunction (ED). Developed through rigorous clinical research, it facilitates the achievement and maintenance of erections sufficient for satisfactory sexual activity by enhancing blood flow to the penis upon sexual stimulation. Its optimized pharmacokinetic profile allows for a rapid onset of action and a favorable side effect spectrum, making it a preferred choice for many patients and clinicians seeking a reliable therapeutic option for managing ED.
Features
- Active ingredient: Avanafil
- Standard dosage strengths: 50 mg, 100 mg, 200 mg
- Rapid onset of action, typically within 15–30 minutes
- High selectivity for PDE5 enzyme, reducing off-target effects
- Administration: Oral tablet, taken as needed
- Duration of effect: Up to 6 hours
Benefits
- Enables reliable and improved erectile function to support sexual activity.
- Offers a rapid onset, allowing for greater spontaneity and reduced planning.
- Provides a sustained window of opportunity, reducing time-related anxiety.
- High PDE5 selectivity may contribute to a lower incidence of certain side effects such as visual disturbances.
- Supports psychological well-being and relationship satisfaction by addressing a key aspect of sexual health.
- Flexible dosing allows for individual titration to optimize efficacy and tolerability.
Common use
Avana is primarily indicated for the treatment of erectile dysfunction (ED) in adult men. It is used to facilitate the achievement and maintenance of an erection when combined with sexual stimulation. It is not indicated for use in women, children, or adolescents. It does not increase libido, offer protection against sexually transmitted infections, or serve as a form of contraception.
Dosage and direction
The recommended starting dose of Avana is 100 mg taken orally approximately 15–30 minutes before anticipated sexual activity. The dosage may be adjusted based on efficacy and tolerability to a maximum of 200 mg or a minimum of 50 mg per day. The tablet should be swallowed whole with a glass of water; it may be taken with or without food, though a high-fat meal may delay the onset of action. The medication should not be taken more than once within a 24-hour period.
Precautions
- Prior to use, a thorough medical evaluation should be conducted to diagnose ED and identify any underlying cardiovascular risk, as sexual activity carries potential cardiac risk.
- Use with caution in patients with anatomical deformation of the penis (e.g., Peyronie’s disease, angulation), or conditions predisposing to priapism (e.g., sickle cell anemia, multiple myeloma, leukemia).
- Caution is advised in patients with severe hepatic or renal impairment; dosage adjustment may be necessary.
- Avoid concurrent consumption of excessive alcohol, as it may increase the risk of orthostatic hypotension and diminish efficacy.
- This medication does not protect against sexually transmitted diseases; appropriate protective measures should be used.
Contraindications
- Concomitant use of any form of organic nitrates (e.g., nitroglycerin, isosorbide mononitrate or dinitrate) or nitric oxide donors in any form.
- History of hypersensitivity to avanafil or any excipients in the formulation.
- Patients for whom sexual activity is inadvisable due to underlying cardiovascular pathology.
- Concomitant use with guanylate cyclase stimulators, such as riociguat.
Possible side effects
Common side effects may include headache, flushing, nasal congestion, nasopharyngitis, and back pain. Less frequently, dyspepsia, dizziness, and visual disturbances (such as changes in color vision) may occur. In rare cases, priapism (a prolonged and painful erection lasting more than 4 hours) may develop, which constitutes a medical emergency requiring immediate treatment to avoid permanent tissue damage. Sudden decrease or loss of hearing or vision has been reported in association with PDE5 inhibitor use, though a causal relationship has not been definitively established.
Drug interaction
- Concomitant use with nitrates is absolutely contraindicated due to the risk of severe hypotension.
- Alpha-blockers (e.g., tamsulosin, doxazosin) may potentiate the blood-pressure-lowering effects; concomitant use should be approached with caution, and a time interval may be recommended.
- Strong CYP3A4 inhibitors (e.g., ketoconazole, ritonavir) can significantly increase avanafil plasma concentrations; a dose reduction to 50 mg is recommended, with a maximum frequency of once every 24 hours.
- Moderate CYP3A4 inhibitors (e.g., erythromycin, aprepitant, fluconazole) may also increase exposure; consider starting with the lower dose.
- Antihypertensive agents may have an additive effect on blood pressure.
Missed dose
Avana is taken on an as-needed basis; it is not intended for daily scheduled dosing. Therefore, the concept of a “missed dose” does not apply. Do not take an extra dose to make up for a missed opportunity.
Overdose
In cases of suspected overdose, standard supportive measures should be employed. Expected effects would be an extension of the drug’s vasodilatory properties, potentially leading to severe hypotension. Management should include maintaining supine position and providing intravenous fluids if necessary. Hemodialysis is not expected to significantly enhance elimination due to high protein binding and extensive metabolism.
Storage
Store at room temperature between 20°C to 25°C (68°F to 77°F), with excursions permitted between 15°C and 30°C (59°F and 86°F). Keep in the original container to protect from light and moisture. Keep out of reach of children and pets. Do not use after the expiration date printed on the packaging.
